Synthesis and Anti-inflammatory Screening of Azetidinyindoles and Thiazolidinylindoles

In search for new leads towards potent anti-inflammatory agents, an array of 5-methoxy-3-(3’-chloro-2’oxo-4-substitutedaryl-1’-azetidinyl)indoles (4a-4e) and 5-methoxy-3-(2’-substitutedaryl-4’-oxo-1’,3’-thiazolidin-3’-yl)indoles (5a-5e) have been synthesized form 5-methoxy-3-substitutedbenzylidenlaminoindoles (3a-3e). Structure of all the synthesized compounds has been analyzed using elemental analysis (C, H, N) and IR, H-NMR Mass spectroscopic techniques. Further, these compounds were subjected to screen for their toxicity profile, anti-inflammatory activity, analgesic activity, ulcerogenic liability and cyclooxygenase assay. Structure activity relationship results of above compounds indicated that 5-methoxy-3-[2’-(p-hydroxy-m-methoxyphenyl)-4’-oxo-1’,3’-thiazolidin-3’-yl]indole (5c) displayed better antiinflammatory activity.
